U.S. health insurers, which pay the bulk of prescription drug costs, act as a buffer between drugmakers and patients, with beneficiaries subject to co-pays and co-insurance based on the prices insurers negotiate with drugmakers.

For years, most countries, including the US, have imposed few or no tariffs on finished drugs, thanks in part to a 1995 World Trade Organisation (WTO) agreement aimed at keeping medicines affordable.

This shift comes after Trump introduced a blanket 10% tariff on other imports last week, as part of a broader effort to bring manufacturing back to the US.

His new "reciprocal" tariffs - including a duty of 104% on goods arriving from China - came into force on Wednesday, intensifying a global trade war and further shaking markets.
Pharmaceutical buyers, so far spared from such measures, are now preparing for what may come next.

The US has typically imported vast quantities of finished medicines from India, Europe and China without buyers paying tariffs - although active pharmaceutical ingredients (APIs), used to make drugs, do face some duties.

Speaking at a fundraiser dinner for his Republican Party on Tuesday, Trump said: "We're going to be announcing very shortly a major tariff on pharmaceuticals. And when they hear that, they will leave China."

He also told reporters on board his Air Force One plane last week that "pharma" tariffs would arrive "at a level that you haven't really seen before", saying these would be announced "in the near future".

In 2024, the US imported $213bn (£168bn) worth of medicines - more than two and a half times the total a decade earlier.

While short on detail, his comments have rattled buyers, especially those relying on Indian imports. India supplies nearly half of all US generics, or cheaper versions of popular drugs, saving the country billions in healthcare costs.
Indian pharma stocks fell sharply on the news. India sends about a third of its $13bn annual pharma exports to the US, which is a key market.

At the moment, Americans pay little or no tax on imports of Indian medicines - compared with the duty of nearly 11% paid by Indians importing American medicines.

Indian drugmakers warn that tariffs would force them to raise prices, which could ultimately drive up US medical bills. While firms like Cipla and Dr Reddy's have US plants, most say moving production is not viable for low-margin generic drugs.

European drugmakers are also on alert. After a high-level meeting between European Commission President Ursula von der Leyen and top pharma firms on Tuesday, the European Federation of Pharmaceutical Industries and Associations (EFPIA) warned that tariffs could shift production away from Europe, and to the US.

The EFPIA, whose members include major pharmaceutical companies such as Bayer, Novartis, and Novo Nordisk - the maker of the star diabetes type 2 drug Ozempic - expressed concerns that rising tariffs could disrupt Europe's role as a key player in global pharmaceutical production.

In 2024, pharmaceuticals were the EU's largest export to the US, worth a reported $127bn (£100bn).

Major companies have urged the EU to act swiftly, seeking policy changes to enhance Europe's competitiveness and prevent a "mass exodus" to the US. They have also expressed concerns about potential EU retaliatory tariffs, which could disrupt supply chains and affect patients on both sides of the Atlantic.

Global pharma giants like GSK and Pfizer operate across several countries, including Ireland and Germany, meaning new tariffs could disrupt multiple parts of the supply chain.

Pharmacists Can Bridge Gaps in the Evolving Esophageal Cancer Treatment Landscape

Pharmacy Times: What are unmet needs in the current therapeutic landscape for metastatic ESCC?

Scott Soefje, PharmD, MBA, BCOP, FCCP, FHOPA: Esophageal cancer is a difficult cancer. It’s one of those that’s hard for people to treat and hard to really get your hands around. It’s not something pharmacists see on a regular basis. It’s also not as common in the U.S. as it is in some other parts of the world.

One of our biggest problems with esophageal cancer is the lack of screening. There’s no reliable way to identify these patients early and get them into treatment at an early stage. So, a lot of times, when patients do show up, the disease is already advanced—or they’re presenting with symptoms like difficulty swallowing or pain when swallowing. By then, the tumor is often already causing significant problems, and that really inhibits our ability to catch the disease early and move toward curative treatment, which is typically surgery. So instead, we’re often left trying to control the disease or palliate symptoms—things like that.

The second big issue is that we can’t look at someone and predict who is at higher risk of relapse. It becomes a bit of a hit-or-miss situation. And unfortunately, a lot of the risk factors that cause squamous cell carcinoma—like prolonged smoking and alcohol use—tend to persist. Many patients don’t give up those habits even after diagnosis, so we’re constantly having to fight both the disease and the ongoing risk factors. It makes it even harder to keep patients moving forward in their journey to beat esophageal cancer.

Then, we’re also dealing with poor overall survival rates. Right now, we’re looking at survival of 20% or less overall. If the disease is locally advanced, survival rates drop to less than 5%. So, it’s really one of those cancers that’s particularly tough to manage.

What’s interesting now is that we do have more treatment options on the table—but we don’t know the optimal sequence. We know surgery is the curative modality. We know other therapies are involved. But how do we best sequence them? Do we give chemotherapy first? Do we combine chemotherapy with immunotherapy upfront? Do we save immunotherapy for relapse? There are a lot of unanswered questions when it comes to treatment sequencing—and that’s becoming a challenge across many tumor types, not just esophageal cancer. So, when you put it all together, we’re left with a real problem: how do we truly manage esophageal cancer across the board?

Pharmacy Times: What are the contributions of pharmacists in managing metastatic ESCC with PD-L1 inhibitors, specifically in relation to formulary management, patient education, and monitoring?

Soefje: For pharmacists involved in managing esophageal cancer within the PD-1 inhibitor space, there are basically three drugs that now have indications: nivolumab, pembrolizumab, and tislelizumab. All three of these have supporting studies showing, to some extent, improvements in progression-free survival, overall survival, and objective response rates.

The problem is, we don’t have head-to-head studies—so we don’t actually know which one is better. That leaves us asking: how do we sequence these drugs? When do we combine them with a chemotherapy backbone, and when do we use them as single agents?

Pharmacists can really make an impact here by helping assess the patient’s performance status. Based on that, which approach makes more sense? Should we go full-blast with chemo-immunotherapy, or should we opt for immunotherapy alone?

Pharmacists also play a crucial role in managing side effects. Immunotherapy agents, in particular, are ideal for pharmacist involvement because of the wide range of potential immune-related endocrinopathies. We can help patients get through those and help providers understand when these side effects typically occur, how to identify them, and how to manage them effectively. So I think this is a great opportunity for pharmacists to step in.

Beyond that, these patients have significant supportive care needs—nutrition, swallowing issues, pain management, and so on. Those are all areas where pharmacists can make a difference.

And then, on top of all of that, the chemotherapy we use in this space tends to be highly emetogenic. Managing nausea and vomiting becomes critical, especially in a patient population that may already be experiencing these symptoms. That’s another area where pharmacists have a clear role in the overall management of these patients.

Pharmacy Times: What are some updates in therapy guidelines and/or emerging treatment data from clinical trials related to PD-L1 therapy in metastatic ESCC that may support pharmacists providing informed, patient-specific recommendations?

Soefje: I think the biggest thing is the inclusion of PD-1 inhibitors in the preferred category in some of the NCCN guidelines. We now have all three of them listed as category one, generally in combination with fluorouracil or a platinum agent. That’s really started to push these PD-1 inhibitors into the front line—it’s becoming clear that we need to be using them.

Then the next step is asking, How do we look at this?" and say, ‘Okay, if we can’t do chemotherapy and immunotherapy—whether due to side effects, performance status, or some other reason—what do we do next? Do we give chemotherapy alone? Do we give immunotherapy alone?"

And then, once patients progress again, immunotherapy is still part of second-line systemic therapies. But by then, we’re usually down to single agents—like taxanes, irinotecan, and others.

So how do we work with our teams to make sure patients are sequenced correctly, that we’re staying within the guidelines, and that we’re optimizing the best care possible based on what the literature is telling us? Those are the key things.

You know, the future is going to get really interesting—especially in the solid tumor realm—and esophageal cancer could definitely be part of that. There are CAR T-cell therapy trials out there. There are other types of drugs being developed. We’re seeing new PD-1 inhibitors under investigation.

At some point, we’re going to have to decide: when is a PD-1 just a PD-1? PD-1 inhibitors are starting to feel like the statins of oncology—there are so many of them, each with their own niche indication. So how do we decide which one to use? Or will we get to a point where we say, “They’re all the same. Let’s pick one as our preferred agent, and that’s the one we’ll use institution-wide”?

I don’t think we’re there yet, but I think we’re heading in that direction in some areas. It’s something pharmacists really need to be on the front lines for—helping the care team put all of this together.

Aurobindo Pharma's arm receives permission from European Commission to market Dyrupeg

Aurobindo Pharma announced that its wholly owned step-down subsidiary, CuraTeQ Biologics s.r.o., has received permission from the European Commission (EC) to market Dyrupeg in the European Union.

The committee for medicinal products (CHMP) of the European Medicines Agency had adopted a positive opinion, recommending the approval of Dyrupeg.

Dyrupeg is a biosimilar medicine containing the active substance pegfilgrastim, intended to reduce the duration of neutropenia and prevent febrile neutropenia in adult cancer patients treated with cytotoxic chemotherapy.

Aurobindo Pharma is principally engaged in the manufacturing and marketing of active pharmaceutical ingredients, generic pharmaceuticals, and related services.

Aurobindo Pharma reported a 10.04% decline in consolidated net profit to Rs 845.57 crore in Q3 FY25, compared to Rs 939.97 crore posted in Q3 FY24. Revenue from operations increased 8.53% year on year (YoY) to Rs 7,978.52 crore in the quarter ended 31 December 2024.

Pharma stocks survive market rout on tariff exemption, but uncertainty continues

LONDON/BENGALURU April 3 (Reuters) - Drugmaker stocks gained a temporary reprieve on Thursday as U.S. President Donald Trump spared pharmaceutical products from reciprocal tariffs, but executives and analysts warned it was premature to celebrate as tariffs were still likely to come.
Trump imposed a 10% tariff on most U.S. imports, as well as much higher levies on dozens of rivals and allies alike, but temporarily exempted some goods, including pharmaceuticals, benefiting major exporters including India, Japan and Ireland.
Shares of U.S. drugmakers AbbVie (ABBV.N), opens new tab and Johnson & Johnson (JNJ.N), opens new tab rose about 2%, defying the broader market drop.
Across continents, shares of Indian and European drugmakers (.SXDP), opens new tab rose, in a sign of relief that pharmaceutical products for now remained out of the crosshairs of the trade wars.
A U.S. official said on Wednesday the president plans separate tariffs targeting the pharma sector.
Trump in his White House Rose Garden announcement once again namechecked the industry, predicting that pharma companies will come "roaring back" to the U.S, and warning if they don't, "they got a big tax to pay".
"The administration has reinforced the need to maintain robust and resilient domestic manufacturing capacity in pharmaceuticals," Stephen Farrelly, global pharma & healthcare lead at ING. "So it is being highlighted as a sector they needs to reshore."
WORRIES PERSIST
Many in the industry predicted the recent uncertainty over tariffs would continue to cast a shadow over drugmakers.
"The only thing that feels certain is more uncertainty," Barclays analyst Emily Field told Reuters.
One source at a European drugmaker said the sense on pharma sector tariffs was: "It's not today, but it's coming."
Trump's executive order listed pharmaceuticals alongside lumber, semiconductors and other sectors that could be subject to investigation under Section 232 of the 1962 U.S. Trade Act.
U.S. manufacturing costs for pharma companies will rise as country-specific tariffs will affect key supplies such as organic chemicals and glassware used to make pharma products, Bernstein analysts said in a note. They calculated an additional $45 billion of import cost risk to the pharma industry.
Jefferies analysts said Biogen (BIIB.O), opens new tab and Amgen (AMGN.O), opens new tab were among the drugmakers with most ex-U.S. exposure, while UBS pointed to AbbVie (ABBV.N), opens new tab and Merck (MRK.N), opens new tab having significant overseas manufacturing.
Medical devices and diagnostics equipment did not appear to be exempted, and shares in companies including GE Healthcare (GEHC.O), opens new tab and DexCom (DXCM.O), opens new tab fell about 6%.
Medical device industry group AdvaMed said the tariffs would likely lead to cuts in research and development spending and threatened the U.S. position as a leader of innovation in the medtech sector.

COPENHAGEN, Denmark — Europe’s pharmaceutical sector is bracing for the potential impact of U.S. tariffs as hopes of an industry-wide exemption by U.S. President Donald Trump fade.

The pharmaceutical industry has until now been exempt from trade levies, but Trump confirmed last week that he would soon impose tariffs on the sector.

Drugmakers are now lobbying the president for a phased approach to duties on imports to the U.S., Reuters reported Tuesday, citing four sources familiar with the discussions. The sources said the levies may not be announced Wednesday but were likely inevitable.

Gradual implementation of tariffs on the sector could reduce the immediate financial hit and allow companies time to relocate their manufacturing stateside. Nevertheless, some firms have warned that the lack of clarity is already having negative secondary effects.

“For us, what is more important is not only the impact of tariffs, it’s also the impact it makes on the market,” Ester Baiget, CEO of Danish biotech firm Novonesis, told CNBC Tuesday.

“When you bring tariffs, it drives uncertainty, and when you [are] uncertain, you pause, you pause innovation, you post launches, you pause investments,” said Baiget, whose firm derives around 30% of its sales from the U.S. but has also been boosting its manufacturing presence in the country.
Denmark is one of Europe’s largest pharma and biotech hubs, home to companies including Wegovy-maker Novo Nordisk and vaccine-producer Bavarian Nordic — both of which have high exposure to the U.S.

Novo Nordisk’s chairman told CNBC last week that the company was not speculating ahead of Trump’s tariff announcement and was instead focused on remaining flexible.

“It doesn’t make a lot of sense to speculate too much,” Chairman Helge Lund told CNBC on the sidelines of the Danish pharmaceutical giant’s Annual General Meeting. “We are laser-focused on what we can impact.”

Still, questions have been asked about how tariffs might impact U.S. sales of Novo’s hugely popular obesity and diabetes treatments — and the implications for U.S. rival and Zepbound-maker Eli Lilly
. Lund would not be pressed on what share of its weight-loss drug sales derived from U.S. plants, instead pointing to the firm’s “very significant” manufacturing set-up in the U.S.

PhRMA used this point in meetings in February and March with the U.S. administration to urge it to consider a staggered tariff rate increase over several years to reflect the time companies need, one of the four sources said.
While the situation remains fluid, the industry is hopeful that any initial tariff announcement on the sector would be below the 25% Trump has repeatedly threatened, the same source said.
The White House did not immediately respond to requests for comment.
NOT A QUICK PROPOSITION
There may be a growing acceptance within the Trump administration that moving drug manufacturing to the U.S. from abroad is not a quick proposition, leading to optimism among some in the industry that the president might consider a gradual ramp-up to 25% tariffs, another of the sources said.
Drugmakers have argued that tariffs could increase the chance of drug shortages and reduce access for patients. Still, Trump has pushed for the fees, arguing that the U.S. needs more drug manufacturing so it does not have to rely on other countries for its supply of medicines.

The Science and Art of Structure-Based Virtual Screening

Structure-based virtual screening has gained momentum again as the high attrition rate at every stage of drug discovery drives the need to explore a greater chemical space. From the Bayesian perspective, its shortcomings as a viable strategy for sustainable hit discovery are discussed, with regard to the prior hit rates of screening libraries and the performance of computational methods. Lessons are shared in selecting virtual hits for experimental validation learned from a series of eight successful campaigns, one of which impacted the discovery of a drug candidate currently in clinical trials.
High-throughput screening (HTS) has been the dominant strategy used in drug discovery to identify chemically tractable matters that modulate the biological activity of selected targets since 1990s. (1) It is both laborious and costly to screen large libraries of compounds. The screening libraries, often proprietary, are inaccessible to biotech companies and academic laboratories. HTS has been complemented by DNA-encoded library (DEL) technology (2,3) and fragment-based lead discovery (FBLD). (4,5) Structure-based virtual screening (VS) aims to select only a few compounds for experimental testing, by predicting affinities of library compounds to the selected target on the basis of their predicted binding modes without requiring a priori knowledge of actives. Over the years, an exponential increase in the applications of VS has been seen in the drug discovery arena. (6−8) The high attrition rate at all stages of drug discovery continues to drive the need for exploration of a greater chemical space. The vast virtual chemical space arising from reaction-based library enumeration and, more recently, AI generative models, brings VS under the spotlight once again. (9−13)

HTS remains the primary tool in hit discovery; this renders VS akin to a lottery, a quick and cheap option to try one’s luck. Given the large number of VS campaigns reported, it is, perhaps, no surprise to see that hits have been identified and some have even been optimized into clinical candidates. (14) In view of the place of serendipity in drug discovery, I focus on whether VS could serve as a sustainable and reliable hit discovery approach, and if so, what are the rate-limiting steps that prevent its general success?

Six structure-based VS campaigns were carried out against the kinases EphB4, (15,16) EphA3, (17) Zap70, (18) Syk, (19) and CK2α, (20) in addition to two against the bromodomains of BRD4 (21) and CREBBP. (22) The hit rates ranged from 9.1% to 75% with a median of 44.4%, by testing around 20 compounds. The IC50 values were in the range between 20 and 0.3 μM, and thus, the hit potency was comparable to that from HTS. The scaffold novelty was checked using SciFinder. The six representative hits were listed in Table 1. At the time that the two VSs were performed against BRD4 and CREBBP, bromodomains were emerging therapeutic targets with few ligands reported, following the landmark publication of JQ1 in 2010. (23) It is noteworthy that the predicted binding modes of four hits were confirmed by crystallography, (16,21,22,24) strongly supporting a causal correlation between their discovery and the computational methods applied. Two chemistry optimizations were conducted on the kinase hits 1 and 2 (16,24) and one on the CREBBP hit 5. (22) Notably, optimization of 1 led to a series of single-digit nanomolar DFG-in/out kinase inhibitors. (24) Compound 7 was selected for preclinical investigations in an array of cell-based assays, gene expression profiling, and PK/PD studies. (25) Encouragingly, compound 7 was reported to be the starting point toward discovery of the drug candidate, RP-6306, which is currently in clinical trials for treatment of various solid tumors. (26) Following its development, Insilico Medicine recently reported the discovery of a novel MYT1 inhibior selective over WEE1. (27) Taken together, one may argue that structure-based VS is indeed a sustainable approach conferring novel chemotypes, with a demonstrated impact on drug discovery.

The quinoline framework and related scaffolds in natural products with anti-Leishmania properties

For almost hundred years, the quinoline heterocycle has been recognized as a privileged pharmacophore in anti-Leishmania agents. Early on, the action of compounds with this scaffold, found in some natural alkaloids, was evidenced against different Leishmania stages and strains. Different structural arrangements containing quinoline framework have been described in different in vitro and in vivo anti-Leishmania alkaloids, namely quinoline proper, isoquinoline, quinolone, among others. In recent years, new quinoline-derivatives isolated from nature have been described, in addition to having carried out in-depth biological studies, in vitro and in vivo, as well as chemical modifications to obtain new leaders. This review updates the state of the art on naturally occurring quinolines and some synthetic derivatives to provide therapeutic tools and strategies to explore new drugs based on this chemosystem for the treatment of leishmaniasis.

NEW YORK/LONDON/LOS ANGELES, March 27 (Reuters) - Some drugmakers are taking the unusual step of sending more medicines by air to the U.S., two executives and two logistics firms said amid fears President Donald Trump's April 2 tariffs announcement could include products made in Europe.
Two European-headquartered drugmakers told Reuters this week they are sending as much of their medicines across the Atlantic as possible over the past several weeks and heard other pharmaceuticals companies were doing the same.
Pharmaceutical products have long been spared from trade wars due to the potential harms.
But Trump's move to increase tariffs on goods from China, including finished drugs and raw ingredients, as well as an early round of tariffs between the U.S. and EU on goods like steel and bourbon, has raised expectations that medicines will join the list.
The U.S. depends on medicines partly produced in Europe that bring in hundreds of billions of dollars in revenue.
Concerns about potential pharma tariffs began before Trump took office in January after he threatened universal import tariffs during his campaign. He has since repeatedly threatened a 25% tariff on pharmaceutical imports and on goods from the EU.

The second executive at a drugmaker said his company was "stockpiling" in the United States, the pharma industry's biggest market worth about $630 billion, a step aimed at easing the immediate blow of tariffs if they are implemented.
The sources spoke on condition of anonymity in order to openly discuss their company's contingency planning for potential tariffs.

While a variety of diseases are driven by aberrant ubiquitination and degradation of critical proteins with protective functions, target protein stabilization (TPS) rather than TPD is emerging as a unique therapeutic modality. Deubiquitinase-targeting chimeras (DUBTACs), a class of heterobifunctional protein stabilizers consisting of deubiquitinase (DUB) and protein-of-interest (POI) targeting ligands conjugated with a linker, can rescue such proteins from aberrant elimination. DUBTACs stabilize the levels of POIs in a DUB-dependent manner, removing ubiquitin from polyubiquitylated and degraded proteins. DUBTACs can induce a new interaction between POI and DUB by forming a POI-DUBTAC-DUB ternary complex. Herein, therapeutic benefits of TPS approaches for human diseases are introduced, and recent advances in developing DUBTACs are summarized. Big Pharma faces heat as India records 307 drug pricing violations

Pharmaceutical companies have repeatedly violated drug pricing regulations by exceeding permitted price hikes, according to a report by the Parliamentary Standing Committee on Chemicals and Fertilisers. As of 6 March 2025, the National Pharmaceutical Pricing Authority (NPPA) recorded 307 cases of violations under Paragraph 20 of the Drugs (Prices Control) Order (DPCO), 2013, which governs price increases for non-scheduled drugs. These violations highlight growing concerns over the unchecked rise in medicine prices and their impact on affordability.

Under the DPCO, 2013, non-scheduled drugs—those not included in the National List of Essential Medicines (NLEM)—are allowed a maximum annual price hike of 10%. Any increase beyond this limit is considered a violation. The NPPA has taken enforcement action against multiple pharmaceutical firms for non-compliance. M/s ANG Lifescience India Limited and M/s Ridley Life Science Private Limited have been blacklisted for failing to adhere to licensing conditions, while M/s Aveo Pharmaceuticals has been issued a Stop Production Order for violating manufacturing standards, according to the report.

Beyond these violations, the parliamentary panel has raised concerns over the pricing strategies of major pharmaceutical companies, including Sun Pharma, Aurobindo Pharma, Dr Reddy’s Laboratories, and Cipla. It has sought a response from the Department of Pharmaceuticals (DoP) on whether these companies’ pricing and distribution practices have made essential medicines unaffordable for the public. The panel has also questioned whether the high cost of life-saving medicines has pushed them beyond the reach of lower-income patients.

A major recommendation in the committee’s eighth report on Demands for Grants (2025-26) is to expand the scope of the NLEM to include more essential and widely used medicines. Currently, only 388 medicines are covered under the NLEM, including around 1,000 formulations, with the NPPA regulating their prices. However, a large number of commonly prescribed medicines remain outside the price control framework, allowing manufacturers to raise prices unchecked. The panel has urged the DoP, NPPA, and the Central Drugs Standard Control Organisation (CDSCO) to regularly review and expand the NLEM to reflect evolving healthcare needs.

“The unchecked increase in medicine prices is a serious concern. The arbitrary exclusion of essential drugs from price control must be addressed immediately to ensure affordability,” the committee stated in its report.

The NPPA has informed the committee that while scheduled drugs under NLEM are strictly regulated, non-scheduled drugs are permitted annual price hikes of up to 10%. However, the committee has cautioned that even these permitted increases have led to a steady rise in drug costs, making critical treatments unaffordable for many.

Antimicrobial resistance (AMR) is recognized as one of the top ten global health threats, posing significant risks not only to human health but also to environmental wellbeing. It is classified as a quintessential One Health challenge. The most concerning pathogens, characterized by multidrug resistance (MDR), extended-drug resistance, and even pan-drug resistance phenotypes, are collectively referred to by the acronym ESCAPE, encompassing Enterococcus faecium, Staphylococcus aureus, Clostridium difficile, Acinetobacter baumannii, Pseudomonas aeruginosa, and Enterobacteriaceae (De Oliveira et al., 2020). Recent developments in this area have shown the potential of heterocyclic compounds to offer more targeted and effective treatments with fewer side effects. Therefore, this Research Topic invited papers that showcased recent progress in medicinal and natural product chemistry, including the isolation and characterization, design, synthesis, and application of heterocyclic compounds as promising anti-infective and anticancer agents.
This Research Topic covers a total of fourteen original research articles investigating ligands spanning broad therapeutic areas like breast/prostate cancer, inflammatory bowel disease, antitubercular ligands, antimicrobials, anticholinergic and antidiabetic. Seven papers focused on exploring heterocyclic compounds from natural products using various techniques such as molecular dynamics, nanoparticles, in silico and GC-MS profiling and quantum analysis while the remaining papers emphasised the use of heterocyclic fragments for various therapeutics activities.
Mashud et al. investigated the potential inhibitory effects of specific compounds present in leaf extract from Mangifera indica on the growth of drug-resistant breast cancer protease. The chemical compounds present in the plant were analysed using molecular modelling techniques, such as molecular docking, molecular dynamics (MD) simulations, quantum mechanics (QM) calculations, and the Absorption, Distribution, Metabolism, Excretion, and Toxicity (ADMET) method, in order to examine three key chemical constituents: quercetin, catechin, and ellagic acid. The ligands underwent extensive testing to determine their effectiveness against the 3w32-overexpressing breast cancer protein. This study used molecular docking tools and molecular dynamic studies identified ligands with strong binding affinity for the breast cancer protein that overexpressed 3w32. The study identified three ligands that not only surpassed the efficacy of the FDA-approved treatment, but also fulfilled the requirements for a possible new inhibitor of breast cancer.
Barik et al. explored the potential use of silver nanoparticles (AgNPs) for treating periodontal infections, synthesized using leaf extract from Azadirachta indica. The eco-friendly green synthesis process utilizes the plant as a natural stabilizer and reducer, facilitating the formation of silver nanoparticles. Various analytical techniques, including transmission electron microscopy (TEM), Fourier-transform infrared spectroscopy (FTIR), Zeta potential analysis, and ultraviolet-visible spectroscopy (UV-Vis), were employed to characterize the AgNPs. The antimicrobial and antioxidant properties of AgNPs were tested to evaluate their effectiveness against periodontal infections. The results demonstrate significant antibacterial and antioxidant activity, inhibiting biofilm formation and bacterial viability. The study has suggested that AgNPs derived from A. indica may serve as a safe, effective, and environmentally friendly alternative to traditional therapies for treating periodontal infections.

Ending of US weight-loss drug shortages prompts compounder complaints

Decision ends pharmacies’ permission to prepare versions of GLP-1 hormone mimics
Novo Nordisk’s semaglutide diabetes and weight loss drugs, Ozempic and Wegovy, are no longer officially in shortage in the US, according to the US Food and Drug Administration (FDA). That means that compounding pharmacies and outsourcing facilities that had been legitimately preparing their own versions of the drugs will no longer be permitted to do so. Groups representing those suppliers have protested the decision, saying supplies are still tight.
Demand for the glucagon-like peptide 1 (GLP-1) hormone-mimicking drugs surged following Wegovy’s approval for weight loss, straining the Danish company’s ability to manufacture enough injector pens. The FDA placed Wegovy and Ozempic on its drug shortage list in March and August 2022, respectively, allowing compounding pharmacies to supply their own versions to meet demand. ‘During a shortage, compounding pharmacists can play a very important role,’ says Erin Fox, a pharmacist and adjunct profession at the University of Utah, US.
The FDA has now said that Novo Nordisk has now developed reserves of finished and semi-finished products ‘such that supply will meet or exceed projected demand’. However, some patients may encounter challenges getting prescriptions filled due issues in the supply chain between Novo Nordisk and the firm’s customers, the agency acknowledged. The FDA is allowing 90 days for outsourcing facilities and 60 days for compounding pharmacies to stop distributing their semaglutide products.
This ramp-off was welcomed by the Alliance for Pharmacy Compounding, which criticised the lack of notice last year when the FDA ended official shortages of Eli Lilly’s tirzepatide GLP-1 products Mounjaro and Zepbound. ‘We were still seeing huge shortages of tirzepatide injections, the commercially available ones, when the FDA removed them from the shortage list,’ says Tenille Davis, chief advocacy officer at the Alliance.
She notes that there were dozens of FDA-registered manufacturers of semaglutide during the two-year period of shortage. ‘Some people wondered if compounding pharmacies exacerbated the shortage by using the semaglutide [active pharmaceutical ingredient, API], but there’s never been a shortage of the API,’ says Davis. ‘The shortage has always been on the finished dosage form – the pens that Novo Nordisk manufactures.’ Compounding pharmacies formulated the drug in vials for syringe injection, ‘just like a patient would get for insulin’, Davis explains. ‘Vials are much easier to make than auto-injector pens,’ says Fox.

The policy recognizes the potential for AI to improve patient care, acknowledges the risks and ethical challenges associated with the use of AI in healthcare settings, and supports the adoption of policies and procedures related to the use of AI.
ASHP's position is that the pharmacy workforce is uniquely positioned to serve as key contributors and domain experts in the advancement of AI in healthcare.
ASHP's Gina Galanou Luchen, director of digital health and data, and Scott Anderson, director of member relations for the section of pharmacy informatics and technology, sat down with MobiHealthNews to discuss the role of AI in health system pharmacy practice.
MobiHealthNews: How is AI being used in health system pharmacy?
Gina Luchen: We’re seeing AI used mostly in the administrative and operational areas in health system pharmacies. Generating communication, assisting with the prior authorization process and analyzing data to find and implement efficiencies in operations are all ways pharmacies are saving time using AI.
In addition, predictive analytics for inventory management and medication demand forecasting are becoming more common, further optimizing operational workflows.
We are seeing slower adoption in the clinical areas, but augmenting practice with AI in clinical areas is the most anticipated opportunity for pharmacy practice. Currently, AI scribes for patient visits are becoming more common, but pharmacists are exploring ways to improve order verification, medication alerts and chart reviews using AI tools.
There is also great opportunity for AI use for drug information, by streamlining and aggregating literature for clinician and patient use.
MHN: Is AI benefiting hospital patients?
Scott Anderson: Patients are increasingly using AI to ask medication-related questions. In some hospitals, AI voice agents are being implemented to help patients quickly learn more about their medications in the inpatient setting, and pharmacists are working behind the scenes to ensure the information is accurate and assessing the use to identify opportunities for more detailed education. Using AI for administrative and operational tasks is also helping pharmacists reach more patients in the hospital by freeing up time for clinical activities.
AI agents assisting with scheduling appointments, helping route patient requests to appropriate providers and improving care logistics also provide an additional benefit for patients who can receive care faster and more efficiently.
MHN: What are the benefits of AI in terms of its impact on how health system pharmacists do their jobs?
Luchen: The use of AI has a strong positive impact on the time a pharmacist is able to spend on clinical activities and patient interactions. The potential of AI to automate low-level tasks, such as initiating prior authorizations and analyzing coding in the electronic health record, allows pharmacists to shift their focus to direct patient care.
With increased implementation of AI tools in clinical practice, there is a reduction in the time it takes to summarize data in the patient chart, collect documentation, locate clinical literature or information, assess a complex medication regimen and predict patient outcomes.
Additionally, the implementation and ongoing maintenance of AI tools provides pharmacists an opportunity to contribute their expertise on the medication use process, increase their digital engagement and develop new skills that ultimately elevate their role and practice.
MHN: Does AI have a future in health system pharmacy in both administrative and clinical areas?
Anderson: Absolutely. Medical and Pharmacy Students Present Innovative Research at Symposium

Each summer between their first and second years of medical school, students are encouraged to step up to the plate and seek out research opportunities to further expand their experience and knowledge base. This past summer more than 100 University of California San Diego School of Medicine and Skaggs School of Pharmacy and Pharmaceutical Sciences students sought out and completed research projects on a wide variety of topics.
Their work was on display at the Summer Research Symposium held on February 18. Each student created a poster to display findings from their various projects. Four students were selected to give oral presentations of their work.
Brian Chavez, second-year medical student, was one student selected for an oral presentation. His project, A Framework for Capturing Acute Physiologic Stress of Baseball Pitching, looked at whether physiological stress markers respond to a simulated pitching bout and could be integrated into performance and recovery assessments.
“The study is important because baseball players, especially pitchers, are at great risk of injury in the upper extremity, specifically at the elbow joint,” said Chavez. “It’s a very common and costly injury. By expanding what has been previously studied in order to better understand these injuries, such as looking at local and systemic physiologic stress markers as opposed to throwing mechanics alone, we can gain a better understanding as to why these injuries happen and maybe provide ways to better prevent them.”
Selecting the subject
As the students are preparing for this summer research project, they are encouraged to pursue projects which align with their interests. Many students create their own projects based on what they are learning in the classroom, while others use personal interests or social issues to seek out existing studies that they can expand on.
Each student is matched with a faculty member who acts as the principal investigator (PI). The PI supervises the student's work, providing guidance, encouragement, and support to help the student develop a question and explore possible answers.
Samuel Ward, P.T., Ph.D., professor in the Departments of Orthopaedics, Radiology and Bioengineering and vice dean for research for the School of Medicine, served as the PI for Chavez’s work.
“Human performance and wellness are becoming increasingly important in biomedical research. On a medical school campus, this is a departure from traditional disease-focused research, but equally important as we envision healthier and longer life spans for our community,” said Ward. “Baseball is an interesting research framework, as the physics of elite performance is captivating for everyone. Our unique ability to layer state-of-the-art biologic and physiologic measurements onto traditional biomechanical measurements, will provide athletes, coaches and medical staff with critical information related to training, injury risk and recovery.”
Both Chavez and Ward are enthusiastic about next steps for this project, which was completed with support from the Wu Tsai Human Performance Alliance, noting that they have received interest from NCAA, minor and major league baseball to potentially incorporate this type of research longitudinally to further identify trends in physiologic markers and how the information may apply in actual game scenarios.
“This project line is already very active and well-funded, with players, coaches, medical and training staff engaged throughout the year,” said Ward. “This is a really unique academic opportunity- athletes and students from all majors on campus can get engaged in the process.”

I’ve always been fascinated by the incredible ways nature and science work together to heal the human body. In my dad’s early medical-practice days back in the 1930s, he in fact had to rely primarily on nature to heal most of the maladies his patients presented with. He simply didn’t have the vast pharmaceutical armamentarium I had, coming along a generation later.
I thought about this as my wife, Tracy, and I began our guided hike deep in the Amazon rainforest (Tapajós National Forest) with colleagues from The Nature Conservancy (TNC). At the outset we had been given a choice: join a group that would cover a longer, more intense, 10-kilometer trek through the dense rainforest, or take a shorter hike with a traditional guide who promised to stop frequently and share the secrets of the trees and plants and insects around us. Tracy and I chose the latter, opting for depth over bragging rights of distance. And it was a decision that expanded the way I think about the powerful connectedness of planetary health with our own human health. Nature is about us.
Our guide grew up in the rainforest; it was his world. We began down a narrow, winding trail, our local guide opening with a disheartening description of how, not too far away, deforestation had so dramatically affected his community of people and destroyed not only his way of life but the richness of the soil, the range of foods he eats, the way he treats illness, and the water he drinks — and everything that we were about to experience. Nxera Pharma Assigns Japan and APAC (ex-China) Rights to Cenerimod for Autoimmune Diseases to Viatris

Tokyo, Japan and Cambridge, UK, 28 February 2025 – Nxera Pharma (“the Company” or “Nxera”; TSE: 4565) has entered an assignment agreement with Viatris Inc. (“Viatris”), a global healthcare company, and Idorsia Pharmaceuticals Ltd (“Idorsia”), regarding the development and commercialization of cenerimod, a clinical-stage immunology candidate for autoimmune diseases, in Japan, South Korea, and certain countries in the Asia-Pacific (APAC) region (excluding China). The agreement was signed concurrently with Nxera’s assignment of its option to these same rights from Idorsia Pharmaceuticals Ltd under its agreement in July 2023 to acquire Idorsia Pharmaceuticals Japan Ltd and Idorsia Pharmaceuticals Korea Co., Ltd.
Nxera will receive an upfront payment of US$10 million from Viatris and is eligible to receive a milestone payment upon regulatory approval of cenerimod in Japan plus royalties on net sales should it be commercialized in the assigned territories. Nxera will pay no option exercise fee or make any other payments to Idorsia in relation to cenerimod.
Viatris will be the exclusive owner of cenerimod rights in Japan and APAC (ex-China). Viatris gained exclusive rights to cenerimod in all other territories from Idorsia through a research and development agreement signed in February 2024 and following this agreement with Nxera, now owns exclusive global rights to the candidate.
Cenerimod, a highly selective, oral sphingosine-1-phosphate receptor 1 (S1P1) modulator, was discovered by Idorsia and is being developed by Viatris and Idorsia for multiple immune disorders, including in Phase 3 trials as a potential first-in-class, oral therapy for systemic lupus erythematosus (SLE).

Emcure Pharma Gets USFDA Form 483 for API Plant

New Delhi, Feb 26 (PTI) Emcure Pharmaceuticals on Wednesday said the US health regulator has issued two observations after inspecting its Pune-based active pharmaceutical ingredients (API) manufacturing facility.
The US Food and Drug Administration (USFDA) conducted a current Good Manufacturing Practices (cGMP) inspection of the plant from February 19-25 this year, the drug firm said in a regulatory filing.
At the conclusion of the inspection, the company received two observations in Form 483, it added.
"We are addressing the observations comprehensively and will respond to the US FDA within the stipulated time frame," Emcure Pharmaceuticals said.
As per the US Food and Drug Administration (USFDA), Form 483 is issued to a firm's management at the conclusion of an inspection when the investigator has observed any conditions that may constitute violations of the Food Drug and Cosmetic (FD&C) Act and related Acts.

FDA issues showcause notice to Aveo for illegal export of opioids

Maharashtra's Food and Drug Administration has issued a show cause notice to a pharmaceutical company here for alleged illegal export of opioids, an official said on Sunday.
Following the Central government's directives on Friday, a joint team of drug inspectors from the Centre and state government raided Aveo Pharmaceuticals, a Palghar-based company allegedly involved in exporting these drugs, the FDA stated in a release.
Aveo Pharmaceuticals, however, has denied the allegations and said it is fully cooperating with the government authorities in the probe.
As per a statement issued by the FDA, the action was taken after the BBC's investigative report highlighted that Tapentadol and Carisoprodol manufactured in India were illegally exported to African countries, where they are misused for recreational purposes.
The company has been served a show cause notice under the Drugs and Cosmetics Act 1940, and officials have assured that all necessary legal measures will be taken without any delay, fear, or favour.
A spokesperson of Aveo Pharmaceuticals said the company has always adhered to the rules and regulations set by various regulatory authorities to manufacture and export its products.

Drug Discovery and Development Faces an Ominous Future from NIH Indirect Costs Rate Cuts

This reduction in the indirect cost (IDC) rate is a considerable cut in financial support for critical overhead and administrative expenses that are essential for health research. Specifically, IDCs are used for facilities such as laboratory space, scientific instruments, research computing infrastructure, safety and regulatory compliance, research core operations, and support staff. They also play a crucial role in sustaining research institutions, and such drastic cuts could lead to the loss of hundreds of millions of dollars for NIH-funded institutions. The implications are profound, threatening to disrupt a wide spectrum of biomedical research, from basic laboratory investigations to clinical trials. Furthermore, the abrupt nature of these cuts leaves institutions with little time to adjust, placing them in immediate financial distress.
The medicinal chemistry community, in particular, will face significant setbacks. NIH-funded academic institutions serve as hubs for early-stage drug discovery research, identifying novel drug targets and pioneering new chemical entities that may ultimately become transformative therapeutics. The pathway from discovery to market often depends on academic institutions partnering with industry collaborations that facilitate clinical trials and the path toward regulatory approval showing how these cuts will have impacts beyond. While the full impact of these IDC rate reductions remains uncertain, it is evident that they will slow the drug development pipeline, increasing both the time and difficulty of bringing new medicines to patients.
At this juncture, it remains unclear how best to navigate these policy changes, despite lawsuits filed and a temporary restraining order. Now, more than ever, we must be united to ensure that scientific progress continues unimpeded and that the discovery and development of life-saving therapies remain a national priority.

The top diplomats of the United States and Russia are making their way over to Saudi Arabia to hold key talks on the war in Ukraine. Trump administration's move to choose Riyadh as the venue for hosting breakthrough US-Russia talks underscores the Kingdom's return to the diplomatic fold from the near pariah state it went under former President Joe Biden's administration.
This will be the first high-level meeting between Washington and Moscow since US President Donald Trump took office, during which top US and Russian diplomats will try to resettle their countries' fractured relations and make a tentative start on trying to end the Ukraine war. They will also prepare for a summit between US President Donald Trump and his Russian counterpart, Vladimir Putin.

Saudi's Rise From Shadows

Mr. Trump's first foreign visit in his first term was to Saudi Arabia, and the Kingdom has welcomed his return to the White House.
One of the possible reasons for Mr. Trump to keep Riyadh close could be a smooth culmination of the Abraham Accords that he initiated in his first term. However, war in Gaza has subsequently got in the way of the deal and may well raise the price that Saudi Arabia will demand for a peace agreement with Israel.
'Big Coup for Saudi'
"It's a big coup for Saudi. The two superpowers come to Riyadh to settle their disagreements," said Ali Shihabi, an advisor to the Saudi government.
"It's quite prestigious and affirms the soft power of the kingdom," he told AFP.
